Analysis of endotoxin levels is a crucial part of quality control in biotech, medtech, and pharmaceutical industries. An accurate and precise analysis can help reduce the risk of adverse effects and ensure high quality and safety of biological products.

Purpose/Benefit:

Endotoxins are components of the outer cell membranes in gram-negative bacteria and when released, can cause serious and sometimes deadly inflammatory reactions in humans and animals. Therefore, it is important to ensure that medical devices, biological and pharmaceutical products and materials are free from endotoxins before use.

Method (what/which methods are used to perform the service):

At RISE, endotoxin levels are analyzed using a kinetic chromogenic Limulus amebocyte lysate (LAL) method according to Eur.Ph.2.6.14 (method D). The LAL test is based on the coagulation reaction of amebocytes from horseshoe crabs, which react with endotoxins. The test is sensitive and specific for endotoxins and can detect very small amounts.

For the analysis, the Endosafe® LAL-cartridge system from Charles River Laboratories, with FDA-licensed cartridges and the Endosafe® Nexgen PTS™ reader is used. The detection limit for the analysis is 0.005 EU/mL.

We can analyze both liquid solutions and solid materials, e.g. medical devices. Solid materials are extracted according to US Pharmacopoeia chapter 161.

Examples of organic substances are additives such as dyes, biocides, surfactants, preservatives, consistency sensors, flame retardants (in, for example, plastics and electronics), plasticizers (phthalates) or other additives in plastics and rubber.

Purpose/Benefit:

The results of the analyzes can be compared against different EU directives (eg. REACH, RoHS and the toy directive, cosmetics regulation) and different chemical lists (SinList, TEDX etc.) to determine whether the material / product contains CMR (carcinogenic, mutagenic, reproductive toxicants) substances, SVHC substances (Substances of Very High Concern) or classified as PBT / vPvB substance (Persistent, bioaccumulative and toxic / very persistent and very bioaccumulative).

Some functional chemicals that have been noted for environmental reasons include PCBs, PAHs, phthalates, perfluorinated substances (PFOA / PFAS), Bisphenol A (BPA) and various volatile and / or odoriferous substances.

Functional chemicals of biological origin can sometimes vary slightly in composition or being sensitive to heating or otherwise affecting the molecular structure. Therefore, quality control and other characterization of biomolecules place high demands on the laboratory.

Organic substances can be detected in various media such as water, tissues, biomull, particles, waste, air or other gas mixtures as well as in different materials and goods. The substances often occur in very low levels.

RISE has several advanced instruments for detecting, characterizing and quantifying organic substances, including:

• UV-VIS-NIR spectrometer with integrating sphere

• Infrared wavelength range (FTIR) spectrometer

• Liquid chromatographs (LC) with various detectors (MS-MS, UV, RI)

• Gas chromatographs (GC) with different detectors (MS, ECD, FID, TCD)

• XRF, surface layer analysis of a material's elemental composition

Tensile testing of metallic materials

A method to determining the mechanical properties such as e.g. yield strength, ultimate strength and elongation under static loading. The testing can be carried out using either flat or cylindrical test specimens that are in line with the relevant standard.

Purpose/Benefit:

The purpose of the test method is to characterize the mechanical performance of metallic materials, which enables to better understand whether the material of interest will withstand the external loads. Moreover, the obtained data can also be used to compare different alloys and/or to study the impact of varying the manufacturing process. In addition, the data can be used to identify parameters of constitutive models to be used in Finite Element Modelling (FEM).

Method (what/which methods are used to perform the service):

The testing is carried out in the lab according to the relevant standards. Flat or cylindrical test specimens are mounted in the testing machine and a load is applied while measuring the resulting strain. Various properties, such as tensile strength and ultimate strength, are obtained from the acquired data. When relevant, the testing can be carried out in an oven (from 300 °C up to 1200 °C) or in a climate chamber (from -100 °C up to +600 °C). Additional measurement techniques, such as Acoustic Emission (AE) and Digital Image Correlation (DIC), can be provided.

Measuring hydrogen gas in aluminium melt by Reduced Pressure Test

Reduced Pressure Test is used to measure  hydrogen gas in aluminium melt. Hydrogen is the only gas soluble in aluminium and may at solidification precipitate as porosity in the cast component. Therefore the level of hydrogen in aluminium should be controlled.

Purpose/Benefit:

The method gives the possibility of measuring gas content in an aluminium melt in order to ensure a melt with as low hydrogen levels as possible.

Hydrogen may precipitate as porosity in aluminium castings, which can affect mechanical properties as well as pressure tightness. Hydrogen results in gas porosity but can also contribute to larger problems in porosity arising at the solidification shrinkage of the material. By measuring hydrogen content in the melt, the foundry may learn what affect the content and be given an indication on how the quality of the casting may be affected. There are research theories that increased oxide content in melt gives more porosity, which means that the method could also be used to evaluate purity in general in the melt. This can then be a tool for evaluating the process in terms of routines for handling the melt, as there is a great risk that the oxide content increases during these processes.

Method (what/which methods are used to perform the service):

Two small samples are taken from the melt, where of one solidifies under vacuum. The vacuum makes the hydrogen expand, which will give a difference in volume related to the sample solidified in normal atmosphere. The samples can be examined by microscopy, but also be used as samples in a following measuring of specific density which is the most common way to evaluate the results. By using an equipment based on the principle of Archimedes a density index may be calculated, which gives the difference in density between the two samples.

Qualiflash - Analysis of fluidity in aluminium melt

Qualiflash® is an equipment that is used to provide a quality index for aluminium melts used for casting. It is important that an aluminium melt is as clean from oxides as possible in order to produce castings of high quality.

Purpose/Benefit:

The method provides the possibility of measuring cleanliness in an aluminium melt in an educational manner. The method should foremost be used as an educational tool to make sure that the internal processes of handling the melt are correct and are working. This can be even more important when using material and recycled material which may contain more oxides due to that it has been remelted several times. This makes the equipment useful for learning about process control and how to set up your melt handling routines.

Large amounts of oxides in aluminium may result in problems as decreased fluidity, mechanical properties and surface treatment properties. It is therefore important that the internal processes at the foundry, when it comes to melt handling, are in place and that relevant staff knows what happens in the different process steps and how the handling is affecting the quality of the melt.

Method (what/which methods are used to perform the service):

The equipment Qualiflash® has a temperature-controlled crucible where the melt is poured. The melt runs through a filter in to a stair-case shaped die. Depending on the cleanliness of the melt a number of steps in the die are filled. More oxides will make the filter plug and less metal will fill the die. By comparing the first reference sample with samples collected after different treatments, an indication on whether the process works and if the melt handling has been successful will be provided.

By reading out of a table a so called Q-index which will give an indication on the fluidity of the melt may be provided.

The measurements are conducted at the foundry together with foundry staff. The requirements are that an operator from RISE participates the first time the equipment is used. The equipment is then possible to rent on a weekly basis if the foundry wants to repeat the measurements to assure the processes.

With hydraulic fluids, power is transferred in many different types of vehicles and work machines, for example in forestry, construction and port operations. The use of hydraulic fluid helps to save energy.
However, hydraulic fluid can inadvertently leak into the environment. Therefore, a Swedish standard (SS 15 54 34) has been drawn up which, among other things, sets environmental requirements for hydraulic oils.

Purpose/Benefit:

To determine whether a hydraulic fluid meets the environmental requirements based on chemical composition based on analysis with FTIR and GCMS.

Method (what/which methods are used to perform the service):

The hydraulic fluid is analyzed with FTIR and GCMS and compared against references on the so-called "SP-list" and other references that may be considered relevant.

When casting aluminium porosity might form in the component due to hydrogen, air or other defects in the material. Measurement of porosity level is an effective way of investigating the amount of porosity in cast aluminium.

Purpose/Benefit:

The method provides the possibility of studying how the material looks inside, for example when analyzing material quality where porosity level is an important factor.

Measurement of porosity level as a continuous process controll is an effective way of controlling the process and may be used as a parameter for quality assurance of the casting process.

Measurement of porosity level gives a relatively fast indication whether the material  meets the requirements regarding amount and size of porosity in a cast component.

Method (what/which methods are used to perform the service):

The method is based on that a sample is cut from a Component. The sample is  mounted in plastic and then subjected to grinding and polishing. The sample is placed in a light optical microscope with a software for image analysis. A rather large amount of pictures from the surface of the sample is analyzed regarding number and size of porosity.

Report from the analysis with porosity level, most common size distribution and possibly pictures from the cross section.

California Technical Bulletin 117-2013 - Requirements, test procedures and apparatus for testing the smolder resistance of materials in upholstered furniture

Purpose/Benefit:

The standard is used to assess the ignitability of individual materials, such as covers or fillings used in upholstered seating, when exposed to an ignition source in the form of a smouldering cigarette.

This test method only evaluates the ignitability of upholstery materials. In order to evaluate the burning behaviour, the furniture needs to be tested in full scale with larger ignition sources.

Since June 25, 2021, all materials in upholstered furniture sold in the United States, should fulfil the requirements in this standard. From June 25, 2022, all upholstered furniture must be labeled in accordance with 16 CFR 1640.

Method (what/which methods are used to perform the service):

The material to be tested is mounted into a test rig together with the applicable standard material. A glowing cigarette is placed in the junction between the seat and the back in the mock-up assembly.

Observations are made to establish the occurrence of any smouldering and/or flaming ignition of the materials. The length of the charring in the assembly is also measured to evaluate compliance.
