Mutagenicity testing

Certain chemical compounds are mutagenic; they can alter DNA and lead to severe diseases like cancer. When working with novel substances, it is essential to assess their mutagenic potential. At RISE, we offer a simplified bacterial reverse mutation test (Ames test), which is a cost-effective initial screening assay to detect mutagenic compounds.

 Purpose/Benefit:

The purpose of the Ames test is to identify whether a substance has mutagenic potential, i.e. the ability to cause genetic changes. The test is used as a first step in the risk assessment of chemical compounds. By using specific bacterial strains, it is possible to quickly and cost-effectively detect the ability of substances to induce mutations.

 Method (what/which methods are used to perform the service):

The bacterial reverse mutation test uses strains of Salmonella typhimurium and/or Escherichia coli to determine whether a compound is mutagenic. The strains carry specific point mutations that prevent them from synthetising an essential amino acid. As a result, they can only grow if that amino acid is present in the growth medium. The test detects reverse mutations that restore the bacteria’s ability to synthetise the amino acid. The revertant bacteria are detected by their ability to grow in medium lacking that amino acid.

At RISE, we offer a simplified version of the method described in the OECD Test Guideline No. 471 (Bacterial Reverse Mutation Test) of the OECD Guidelines for the Testing of Chemicals. This version uses two S. typhimurium strains: TA98, which carries a frameshift mutation, and TA100, which carries a base-pair substitution. Both mutations are in the histidine operon, making the strains unable to produce histidine.

The test involves exposing the strains to six concentrations of the test compound, both with and without a metabolic activator. The bacteria are then incubated in a histidine-free medium. A dose-dependent increase in the number of revertant colonies indicates that the compound is mutagenic under the test conditions.

Emergency communications leaves Rakel and takes the next secure step

The alarm sounds – and the transmission must not fail. Ensuring that vital emergency communications information gets through can mean the difference between life and death. As the current communication system is replaced by a new one based on 5G, the testing methods for a more robust system are already in place.

The current communication system for emergency services, Rakel, has been in use since 2006 and is well established. However, the system focuses on voice communication and does not have modern support for sending messages via apps or video from drones, which is what emergency services need today.

Time for systems that can transmit data and video

That's why a new system, Swen, is being introduced that can transmit data and video as well as voice. Swen stands for The Swedish Emergency Network.

"With systems that are supposed to be very reliable, you can't have a phase where they don't work. Everything has to work right away, so you have to supplement that with testing to find any bugs before you put the systems into service. "Everything you use to give the ambulance a route, for example, has to be compatible – such as phones, drones and apps," says Peter Janevik, CEO of the AstaZero test and demonstration facility at RISE.

Parallel systems

Rakel's current users will also have to adapt their own systems and analyse what they want to get out of the new communication system.

"This requires new standards and new ways of thinking. For example, how to configure base stations for 5G, or how to ensure that a device from one company can communicate with devices from other companies," says Peter Janevik.

In the timetable from MSB, the Swedish Civil Defence Agency, 2025-2027 is dedicated to construction, testing and evaluation. In 2028, Swen will become operational, while Rakel will continue to be the basis for operations. After evaluation, there will be a transition period of two years during which Rakel and Swen will be used in parallel. The aim is that everyone currently using Rakel will have switched to Swen by 2030.

The new system uses the commercial mobile networks, and one of the MSB's requirements is that it must be possible to prioritise traffic for emergency communications in the event of major events, major accidents or terrorist attacks.

Security is central to the procurement of new equipment, and the MSB imposes strict suitability requirements on external partners.

Other requirements for the new system are that the mobile terminals must be able to withstand the Nordic weather conditions and that there must be physical buttons for alarm calls.

This provides a unique opportunity for police forces in Europe, for example, to communicate with each other.

Possible cooperation with other countries

Swen will provide new opportunities for police and emergency services in different countries to work together.

Other sectors, such as logistics companies and ports, are also expected to want to use systems based on similar technology.

Having the expertise to test

Before new equipment and applications can be approved by the MSB, they need to be tested. RISE contributes to this through its expertise at AstaZero, but also through other test and demonstration facilities within RISE.

"The EMC (Electromagnetic Compatibility) facility provides component-level test facilities where you can see if the equipment meets the requirements. And at AstaZero's test track outside Gothenburg, we can then test that the entire networks work as intended, because we can configure our networks as if they were MSB's networks," says Peter Janevik.

Rakel and Swen – two systems for mission critical communication

  • The term Mission Critical Communication (MCC) is used when it is important that a complete message is delivered.
  • The current communication system, Rakel, and the upcoming system, Swen, are two types of Mission Critical Communication.
  • Rakel, which was operational from 2006 to 2010, focuses on voice and is based on the so-called Tetra technology, a standard for digital radio communication developed in the 1990s. This was before the era of smartphones, when data speeds were slower than today.
  • Swen is based on 5G technology and handles voice, data and video.
  • Rakel is used by more than 650 organisations and authorities in Sweden, including the police, defence, emergency services, public transport, energy companies, municipalities, regions and county councils.
  • The aim is for all organisations currently using Rakel to switch to the new Swen system by 2030.

Empowering a Pan-European Network to Counter Hybrid Threats

Objectives: 

EU-HYBNET (Empowering a Pan-European Network to Counter Hybrid Threats) project aims to empower its network to fight against hybrid threats by proliferating knowledge and facilitating cooperation between industry, practitioners and academia, and by providing advanced solutions for network collaboration and delivering recommendations for training, standardization and industrialization of cutting-edge innovations. 

The main project objectives are: 

  1. To enrich the existing network countering hybrid threats and ensure long term sustainability. 
  2. To define common requirements that can fill knowledge gaps, deal with performance needs, and enhance capabilities of research, innovation and training endeavours concerning hybrid threats. 
  3. To monitor developments in research and innovation activities as applied to hybrid threats.
  4. To indicate priorities for innovation uptake and industrialisation and to determine priorities for standardisation for empowering the Pan-European network to effectively counter hybrid threats.
  5. To establish conditions for enhanced interaction with practitioners, industry and academia for meaningful dialogue and for increasing membership in the network.
  6. To foster capacity building and knowledge exchange on countering hybrid threats.
  7. To create a basis for establishing effective synergies with existing European, national and sub-national networks of practitioners and other actors countering hybrid threats. 

Concept and approach 

To achieve the objectives of the project, we will apply an iterative approach, consisting of three full scale project cycles in three core activity areas:

  1. research (incl. training), 
  2. innovation monitoring, and 
  3. recommendations for standardisation and innovation uptake (including industrialisation).

 Additionally, in a concluding fourth cycle, the project will collect results from each of the previous three cycles, draw conclusions, and make suitable recommendations.

 Purpose/Benefit:

UNECE R155 and R156 are two important regulations aimed at improving cybersecurity and the management of software updates in vehicles.

UNECE R155 focuses on protecting vehicles against cyberattacks by requiring vehicle manufacturers to implement robust cybersecurity measures. The regulation came into effect in January 2021 and became mandatory for all new vehicles produced from July 2024.

UNECE R156 focuses on ensuring secure and reliable software updates for vehicles. This regulation also came into effect in January 2021 and follows the same timelines as R155, with mandatory application for all new vehicles since July 2024.

RISE is approved by the Swedish Transport Agency as a Technical service and perform testing according to the UNECE R155 and R156 regulations. A Technical service is an organisation that has been approved as a testing laboratory and registered in the EU and ECE to conduct tests in connection with type approvals.

 Method (what/which methods are used to perform the service):

The ISO/SAE 21434 standard is used to ensure cybersecurity requirements in electrical/electronic (E/E) systems intended to implement functionality in road vehicles. ISO/SAE 21434 sets overall requirements as well as requirements related to the different parts of the product development lifecycle. 

Examples of activities required include vulnerability analyses and risk assessments, as well as lifecycle-specific activities such as requirement management, testing, and static code analysis.

ISO 24089 is an international standard introduced in early 2023 that focuses on software updates for road vehicles. This standard specifies requirements and recommendations to ensure that software updates are carried out in a secure and reliable manner.

  • Consulting and Guidance: Help with interpreting and understanding the requirements of UNECE R155/R156 or the standards ISO/SAE 21434 and ISO 24089.
  • Testing: Conducting accredited tests against standards to ensure that the vehicle meets cybersecurity and update requirements.
  • Initial assessment: Review and assessment (CoP) of manufacturers' quality management systems in the area of type-approval.
  • Certification: Testing confirming that the vehicle meets UNECE R155/R156.

Refrigerated cargo units (reefers) are an integral part of sea-borne transport, but at the same time, reefer electrical faults are also a common cause of fires. Fires on ro-ro ships pose significant dangers and extinguishing them may often be difficult. Consequently, monitoring ship connected reefers could be an effective approach to fire prevention. MoRe will develop a solution for reefer monitoring, including both hardware and a proposed human-machine interface for the engine control room. This work is a continuation of studies performed in the LASH FIRE project. The finished solution will be installed and evaluated for performance on the Stena Scandinavica. The project group brings together a mix of expertise in engineering, human-centered design and ship/cargo operations, catering for end-results with a high level of practical applicability. These results will be communicated in ways that maximize the chances for industry uptake.
